Single Stroke needs one shot that knocks the bucket off a Risen Grenadier. The bucket is the rusted pail held at the creature's centre; its journal entry says that is where it throws its grenades from.
Any firearm works, and the Pistol from Chapter I is enough. Risen Grenadiers appear in ten regions (24 spawners), starting with one on The Battlefield and six in the Valley of Ashes, so you meet them early. With 500 Health at placement level 1 they won't die before you hit the bucket. Aim for the bucket rather than the head, and keep your distance: their grenades are thrown a long way.
